HSV-Ⅰ感染后人小胶质细胞MX1蛋白表达变化及其作用初探The preliminary exploration of the change of myxovirus-resistant 1protein expression in human microglia after herpes simplex virus-I infection and its action

摘要(Abstract):
目的观察人小胶质细胞在单纯疱疹病毒Ⅰ型(HSV-Ⅰ)感染后抗黏液病毒(Mx1)蛋白表达及干扰素-α(IFN-α)、IFN-β表达变化,初步探讨Mx1蛋白在单纯疱疹病毒(HSV)性脑炎中的可能抗病毒作用。方法应用流式细胞技术检测人小胶质细胞感染HSV-Ⅰ后4、12、48hMx1蛋白的相对表达,采用酶联免疫吸附测定(ELISA)方法检测细胞培养上清液中IFN-α、IFN-β的表达。结果 HSV-Ⅰ实验组各时间点人小胶质细胞Mx1蛋白表达、IFN-α及IFN-β表达均高于正常对照组(均P<0.01);HSV-Ⅰ实验组48h内人小胶质细胞Mx1蛋白表达及IFN-α、IFN-β表达随时间延长升高,且Mx1蛋白表达与IFN-α、IFN-β表达均存在正相关(r=0.632,P<0.01;r=0.714,P<0.01)。结论 HSV-Ⅰ感染可引起体外培养的人小胶质细胞Mx1蛋白表达上调及IFN-α、IFN-β分泌增加,Mx1蛋白可能参与了中枢神经系统抗HSV-Ⅰ过程。
